Being a lignocellulosic feedstock, Napier grass requires effective pretreatments to improve its enzymatic digestibility and fermentability. In this study, Napier grass was subjected to a mild ethanol-based organosolv pretreatment, where HCl and H2O2 were employed as catalysts to promote delignification and hydrolysis of hemicellulose. Experimental design was utilized to optimize the pretreatment condition, and the three main factors investigated were reaction temperature, H2O2 level, and HCl concentration. The ranges of temperature, H2O2 level, and HCl concentration were 70-90 °C, 0-10 wt%, and 3-9 wt%, respectively. The results show that at 90 °C, 5% H2O2, and 9% HCl, the glucan conversion rate could reach 96.7%, with a glucose yield of 79.7%. The fermentability of the pretreated feedstock was demonstrated by ethanol production through simultaneous saccharification and fermentation. The pretreated Napier grass produced 20.9 g/L of ethanol, corresponding to a yield 55.3%. The untreated Napier grass, on the other hand, produced much less ethanol; the ethanol concentration was 9.9 g/L.
