Effect of Chinese Herb Compound Prescription A001 on Antigen Specific T Helper Cell in Mouse Allergic Asthma Model

碩士 === 大同大學 === 生物工程學系(所) === 106 === The major components of traditional Chinese medicine combination A001 are Astragalus membranaceus, Perillae Fructus, and American ginseng. It has been reported that Astragalus membranaceus, Perillae Fructus, and American ginseng modulate the T helper 1 (Th1)/Th2 balance and attenuate asthmatic responses in a murine asthma model. The aim of the present work is to investigate the immune modulatory effects of A001 in a murine asthma model. The results showed that the high dose A001 (LH) significantly suppressed the serum production of ovalbumin-specific immunoglobulin E (OVA-IgE), whereas the lower two dose groups tended to reduce the level of OVA-IgE. All three dose groups (LH, LM and LL) tended to inhibit the production of interleukin (IL)-4 of splenocytes followed by OVA stimulation, whereas the production of interferon (IFN)-γ was not altered. Furthermore, LH and LM significantly reduced the percentage of eosinophils in bronchoalveolar lavage fluid (BALF) and LH suppressed the level of IL-5 in BALF. All the three groups attenuated the relative increase ratio followed by methacholine challenge. Taken together, the present study demonstrated A001 markedly suppressed the functional activity of antigen-specific Th2 cells. These results provide critical insights of A001 on both humoral and cell-mediated immunity in asthma patients.
