碩士 === 明志科技大學 === 化學工程系碩士班 === 107 === Abstract
Daphne genkwa as a common traditional Chinese herbal medicine,
its flower has the functions of water removal and clear sputum and
has been used in Taiwan for a long time. It is often used to
revitalize the body. Many scholars in modern times have studied in
depth various aspects. Most of its extraction uses traditional
solvent extraction as research, At present, no literature has
reported the use of supercritical carbon dioxide to extract Daphne
genkwa, and to explore its active ingredients. This study explored
the extraction conditions in the following conditions: pressure at
200 bar, 250 bar, 300 bar, temperature at 313 K, 323 K, 333 K, carbon
dioxide flow rate at 2 L/min, 3 L/min to influence the rate of
extraction.
The sample was dissolved in methanol and was analyzed by using
a MetaChem C18-A(250×4.6mm,5μm) column with a gradient elution
composed of 5% acetic acid aqueous solution and methanol(40/60,
v/v). Flow rate 1mL/min and chromatographic temperature 25 °C tube.
Under this condition, the peaks of the apigenin was identified by
comparing the retention time with that of the standard reagent.
Typical retention time of Apigenin was 7min. The best operation
coudition of extraction Apigenin is in pressure 250 bar, temperature
333 K, and CO2 flow rate kept at 2L/min.
