Summary:碩士 === 國立中興大學 === 物理學系 === 93 === In this work, silicon masters with various patterns are fabricated by anisotropic etching. And a soft material, poly(dimethylsiloxane), is used to replicate several polymer microstructures from these silicon masters. Scanning electron microscope (SEM) and scanning probe microscope (SPM) observations verify that the polymer microstructures are generated without defects or distortion. In addition, this polymer serves as a biological substrate because it has weaker adhesion force to tip. Moreover, the adhesion forces are measured in buffer and various solutions on poly(dimethlsiloxane) and mica surfaces respectively. The observation indicates that the smaller resistance exists when AFM operates in liquid than that in the air. Finally, the conformations of DNAs and proteins are observed with with AFM in liquid.
