At first, selected enzyme should be able to hydrolyze isolated soy protein (ISP) to increase its immunomodulating activity. ISP hydrolysates (ISPH) of six enzymes were prepared, respectively. The result showed hydrolysis of ISP with Pepsin for 4 h to obtain the hydrolysate (P-ISPH4h) had the highest relative phagocytosis as well as maintained the best yield of 1.45 mg N/mL. Finally, based on flow cytometry analysis, 1 kDa MWCO permeate showed increase of both M1and M2 macrophage proliferation and resulted in higher phagocytosis activity, anti-inflammation and protective effect from lipopolysaccharide (LPS) by affecting M1/M2 phenotype and favoring M2 polarization.Without LPS, only IL-10 significantly increased. With LPS, there was a tendency to decrease IL-6 and IL-10 as the dosage of the permeate increased. The ratio of IL-6/IL-10 was also decreased, which revealed the anti-inflammation potential of the permeate. Furthermore, IL-4 was not affected by adding the permeate regardless of LPS, which result in stimulating differentiation of macrophage into M2c by IL-10. The above results indicated that the immunomodulating activity of 1 kDa MWCO permeate may be due to polarizing phenotype M1/M2 and prefering M2 macrophage.
