The G. lucidium mycelium fermented cereals contained the bioactive substances of -glucan, and triterpene. Various extracts by 100oC water extraction followed by ethanol precipitation (A), 70oC water extraction (B), 70oC water extraction followed by ethanol fractionation (C), 95% ethanol extraction (D), 50% ethanol extraction (E), 10% ethanol extraction (F), and 10% ethanol extraction using microwave (G) of G. lucidium fermented product were prepared and the contents of -glucan, triterpene and total phenolics were analyzed. The triterpene and -glucan contents for the 7 extracts were 20.32 - 62.00 mg/g, and 3.39 - 44.74 %, respectively, with the extract A and D being the highest triterpene content, and the extract A the highest -glucan content. Total phenolics contents for all extracts were 22.37-33.87 mg/g gallic acid eguivalent, with the extract C being the highest. Median effective concentration (EC50) ferric-ion reduction for the extracts A to G were 4.76 - 7.83 mg/mL, respectively. Median inhibitory concentration for DPPH scavenging activity for the extracts A to G were 3.94 - 9.63 mg/mL. The relationship between glucan contents vs. DPPH scavenging activity and glucan contenta vs. ABTS scavenging activity were strong, with correlation coefficient values (r) being -0.799*, -0.985*, respectively. The relationship between phenolics contents vs. DPPH scavenging activity and phenolics contents vs. ABTS scavenging activity were strong, with correlation coefficient values (r) being 0.832, 0.916, respectively. Cell viability of RAW264.7 cells after treatment with H2O2 (0.5 mM) were 78%. The cell viability can be increased to 84.24 - 94.49 % by adding the 7 tested extracts in medium, with the extract A and F being the highest. A significant increase in catalase (CAT) activity was observed in the extract A and extract F treated group. The NO production in RAW264.7 cells treated with 100 μg/mL extract D (22.14 μM) was significantly lower than that of LPS-induced control (35.10 μM). The relationship between triterpene contents and NO inhibition rate was strong (r = 0.922*).
